Modulight Spotlights: LASER-SHARP RESEARCH – September 2023

Modulight Spotlights: LASER-SHARP RESEARCH – September 2023 Head & neck cancer is world’s sixth most common cancer, with increasing incidence and poor survival. A new study by P. Enzian & R. Rahmanzadeh at University of Lübeck, studied combining chemotherapy with photochemical internalization for more effective H&N cancer treatment in vitro. Photochemical internalization using photosensitizer fimaporfin, activated with Modulight’s ML6600 laser, was shown to increase drug’s efficacy by means of better drug delivery inside the cells. As a result, tumor cell death was achieved using 20-fold Continue reading →

Towards Photodynamic Image-Guided Surgery of Head and Neck Tumors: Photodynamic Priming Improves Delivery and Diagnostic Accuracy of Cetuximab-IRDye800CW

Published in: Frontiers in Oncology Authors: Chanda Bhandari, John Fakhry, Menitte Eroy, Jane Junghwa Song, Kimberley Samkoe, Tayyaba Hasan, Kenneth Hoyt, Girgis Obaid    Published in: Frontiers in Oncology Authors: Chanda Bhandari, John Fakhry, Menitte Eroy, Jane Junghwa Song, Kimberley Samkoe, Tayyaba Hasan, Kenneth Hoyt, Girgis Obaid   The study showed that photodynamic priming (subtherapeutic doses of light using ML6500) of tumor microenvironment with nanoliposomal BPD is highly beneficial for delivery of antibodies (Cet-IR800) to H&N tumor in mice. Priming significantly improved the tumor accumulation of Cet-IR800 and diagnostic accuracy of tumor detection, as monitored with fluroescence imaging.   Read the article here

Rapid Depletion of Intratumoral Regulatory T cells Induces Synchronized CD8 T and NK Cell Activation and IFN-γ-dependent Tumor Vessel Regression

Published in: Cancer Research Authors: Yutaka Kurebayashi, Colleen P. Olkowski, Kelly C. Lane, Olga V. Vasalatiy, Biying C. Xu, Ryuhei Okada, Aki Furusawa, Peter L. Choyke, Hisataka Kobayashi, Noriko Sato    Published in: Cancer Research Authors: Yutaka Kurebayashi, Colleen P. Olkowski, Kelly C. Lane, Olga V. Vasalatiy, Biying C. Xu, Ryuhei Okada, Aki Furusawa, Peter L. Choyke, Hisataka Kobayashi, Noriko Sato   Tumor-infiltrating regulatory T cells (Tregs) were selectively depleted using anti-CD25 antibody fragment mediated NIR-PIT with ML7710 (50 J/cm2) in tumor-bearing mice. This induced transient IFN-γ expression in CD8 T and NK cells, which lead to efficient and rapid tumor vessel regression, intratumoral ischemia, and tumor necrosis/apoptosis and growth suppression. These results indicate the pivotal Continue reading →

Irradiance controls photodynamic efficacy and tissue heating in experimental tumours: implication for interstitial PDT of locally advanced cancer

Published in: British Journal of Cancer Authors: Gal Shafirstein, David A. Bellnier, Emily Oakley, Sasheen Hamilton, Michael Habitzruther, Lawrence Tworek, Alan Hutson, Joseph A. Spernyak, Sandra Sexton, Leslie Curtin, Steven G. Turowski, Hassan Arshad, Barbara Henderson    Published in: British Journal of Cancer Authors: Gal Shafirstein, David A. Bellnier, Emily Oakley, Sasheen Hamilton, Michael Habitzruther, Lawrence Tworek, Alan Hutson, Joseph A. Spernyak, Sandra Sexton, Leslie Curtin, Steven G. Turowski, Hassan Arshad, Barbara Henderson   Currently delivered light dose (J/cm2) is the principal parameter guiding interstitial photodynamic therapy (I-PDT) of refractory locally advanced cancer. The aim of this study was to investigate the impact of light dose rate (irradiance, mW/cm2) and associated heating on tumour response and cure. Laser was used to activate Continue reading →

Disulfonated tetraphenyl chlorin (TPCS2a)-induced photochemical internalisation of bleomycin in patients with solid malignancies: a phase 1, dose-escalation, first-in-man trial

Published in: Lancet Oncology Authors: Ahmed A Sultan, Waseem Jerjes, Kristian Berg, Anders Høgset, Charles A Mosse, Rifat Hamoudi, Zaid Hamdoon, Celia Simeon, Dawn Carnell, Martin Forster, Colin Hopper    Published in: Lancet Oncology Authors: Ahmed A Sultan, Waseem Jerjes, Kristian Berg, Anders Høgset, Charles A Mosse, Rifat Hamoudi, Zaid Hamdoon, Celia Simeon, Dawn Carnell, Martin Forster, Colin Hopper   Photochemical internalization with bleomycin was studied in a Phase 1, first-in-man trial for local recurrent, advanced, or metastatic H&N cancer. Modulight laser was used for activation of TPCS2a photosensitizer at the surface of the target tumor. 12 patients completed the 3-month follow-up, with maximum tolerated dose of TPCS2a being established to 1.0 mg/kg.   Read Continue reading →

Yet, common PS lack tumor Continue reading →
