The study investigated photothermal (ICG) and photo-oxidative (verteporfin) activation of liposomes for anticancer drug delivery. Surprisingly, photoactivation of paclitaxel-loaded liposomes did not release paclitaxel with neither photothermal nor photo-oxidative release even at highest tested light doses. ML8500 was used for photo-oxidative release (689 nm) while ML6600 with MLAKIT for photothermal release (808 nm). Molecular dynamic simulations showed that paclitaxel remains trapped within oxidized lipids after liposomal disruption. While results show release of lipophilic drugs might not be successful, verteporfin-containing liposomes successfully released hydrophilic calcein.
